How the Agilent 1100 HPLC Works

Step 1: Solvent Degassing

The 4-channel vacuum degasser (G1322S) continuously removes dissolved gases from up to 4 solvents at up to 10 mL/min total flow, ensuring a stable, bubble-free mobile phase for excellent baseline performance.

Step 2: Solvent Delivery

The quaternary pump (G1311A) uses a high-speed proportioning valve and pre-configured vacuum degasser for low-pressure mixing of up to 4 solvents. The binary pump (G1312A) uses dual floating pistons in series for pulse-free high-pressure binary mixing from 0.001 to 5 mL/min.

Step 3: Sample Injection

The G1329A autosampler draws 0.1–100 µL aliquots from up to 100 sample vials and injects them into the mobile phase stream with minimal carryover (<0.1% typical) and high injection precision for accurate quantification.

Step 4: Column Separation

The sample passes through the temperature-controlled column compartment (G1316A), which uses Peltier heating/cooling to maintain column temperature from 10°C below ambient to 80°C. Stable temperature ensures consistent retention times.

Step 5: Detection & Data Acquisition

Separated compounds enter the detector — DAD, VWD, FLD, or RID. The DAD (G1315A) simultaneously collects full UV-Vis spectra across 1024 diodes (190–950 nm), allowing peak purity assessment and spectral confirmation. ChemStation or OpenLab software processes data and generates compliant reports.
